The station serves the 1970s New Town of Glenrothes, the centre of which is around two miles away by road. A frequent bus service connects the station with the town.

The station was opened by the Edinburgh and Northern Railway in 1847 on what eventually became the North British Railway's line from Edinburgh to Aberdeen via Dundee.

Re-modelled in 2006-2007, at which time the platforms were lengthened, the station is bright and modern, with a ramped footbridge and a lift serving the southbound platform. The new station building is located on the northbound platform on the site of the sidings and shed which were in existence into the 2000s.

Markinch was the home of LNER Class A4 steam locomotive number 60009 Union of South Africa for several years. It was kept in the shed until until 1994. The wooden shed was destroyed by fire in 2003.

The original Category B Listed LinkExternal link Italianate style station building survives, but is not in railway use.

The station sees around 250,000 passenger journeys per year.
